Tuberculosis Bovine
| Species |
Cattle, horses, cervids, pigs, sheep, goats, carnivores. |
|---|---|
| Composition |
Purified tuberculin for mammals is presented as a transparent solution, ranging in color from pale yellow to light brown, without inclusions. 1 ml contains 20,000 IU of purified protein derivative from Mycobacterium bovis strain N8 with a molecular weight of 300-5 kDa, cultured in liquid medium, inactivated, ultrafiltered, and stabilized. |
| Indications |
For in vivo allergic diagnosis of bovine-type tuberculosis. |
| Dosage and method of administration |
Tuberculin is administered intradermally at a dose of 0.2 ml. Before administration, the injection site is clipped, except for goats, sheep, and carnivores. A skin fold is grasped vertically, measured with a cutimeter, recorded, and disinfected with 70% ethanol. The injection is performed immediately at the center of the clipped area, administering 0.2 ml intradermally. A tuberculin needle attached to a 1 ml sterile graduated syringe is used, inserted at an angle outward into the skin. Proper inoculation is confirmed by the formation of a small swelling the size of a pea. Cattle, cows (regardless of gestation stage), bovine young stock over six months, and deer: Administer intradermally in the middle third of the neck. For bulls, inject in the caudal fold. Reaction readings are taken 72 hours later by examining, palpating, and measuring the skin fold, with the results recorded. Negative reaction: A swelling of less than 2 mm without edema, necrosis, exudate, or lymphatic inflammation. Doubtful reaction: A swelling of 2-4 mm without edema, necrosis, exudate, or lymphatic inflammation. Positive reaction: A skin fold over 4 mm with edema, exudate, necrosis, and inflamed, painful lymph nodes. Positive animals are subjected to a comparative test involving simultaneous inoculation of bovine and avian tuberculin on opposite sides of the neck using separate needles and syringes. Results are read after 72 hours and interpreted as in the single test. Positive comparative reaction: A more pronounced skin fold with mammalian tuberculin than avian tuberculin by at least 4 mm, or inflammation at the injection site. Doubtful reaction: Mammalian tuberculin reaction 1-4 mm larger than avian tuberculin, without inflammation. Negative reaction: Mammalian tuberculin reaction equal to or less pronounced than avian tuberculin. Pigs: Inject 0.2 ml intradermally on the dorsal side of the ear, 2-3 cm from its base, or for piglets aged 2-3 months, in the lumbar region, 5-8 cm from the spine, using an intradermal device. Reaction readings are taken after 48 hours. Goats and sheep: Inject 0.2 ml in the lower eyelid, 1.5-2 cm from the free edge. Read the reaction after 48 hours by comparing the state of both eyes. Intrathigh administration using a needle-free device is also permitted, with reactions read after 48 hours. A positive reaction is marked by significant swelling. Carnivores: Inject 0.1 ml in the upper eyelid. Read the reaction after 48 hours by comparing both eyes. A positive reaction is indicated by inflammatory processes in the injected eye. Horses and other equines: Perform an ophthalmic test by instilling 3-5 drops of purified tuberculin for mammals into the conjunctival sac using a pipette or needleless syringe. Read the reaction at 6, 9, 12, and 24 hours by examining the conjunctival sac. Positive reaction: Characterized by purulent discharge, hyperemia, and conjunctival edema. |
